สถาบันการแพทย์จักรีนฤบดินทร์ คณะแพทยศาสตร์โรงพยาบาลรามาธิบดี มหาวิทยาลัยมหิดล เกิดจากพระราชปรารภและพระราชดำริของพระบาทสมเด็จพระเจ้าอยู่หัว ที่มีพระราชประสงค์ให้มีสถานพยาบาลหรือโรงพยาบาลของรัฐขนาดใหญ่ระดับโรงเรียนแพทย์ขึ้นที่จังหวัดสมุทรปราการ เพื่อให้การดูแลและให้บริการตรวจรักษาประชาชนในจังหวัดสมุทรปราการ ซึ่งประชาชนส่วนใหญ่มาจากต่างจังหวัด เพื่อประกอบอาชีพในโรงงานอุตสาหกรรมที่มีอยู่เป็นจำนวนมาก ตลอดจนถึงประชาชนในบริเวณใกล้เคียงรวมถึงจังหวัดชายฝั่งตะวันออก ตั้งแต่จังหวัดชลบุรี ระยอง จันทบุรี ตราด ฉะเชิงเทรา ปราจีนบุรี และสระแก้ว อีกด้วย เนื่องจากในปัจจุบันโรงพยาบาลส่วนใหญ่ในจังหวัดสมุทรปราการเป็นโรงพยาบาลเอกชน ประชาชนซึ่งมีรายได้น้อยไม่สามารถเข้าถึงบริการของโรงพยาบาลเอกชนเหล่านี้ได้
